Ac-Val-Lys-Leu-Gln-ACC is an Mpro peptide substrate for both SARS-CoV-2 and SARS-CoV main protease (Mpro, also known as 3CLpro). A HyCoSuL (Hybrid Combinatorial Substrate Library) approach determined that this sequence was the most preferred substrate containing all natural/proteinogenic amino among those tested (KM = 228.4 ± 9.9 µM, Kcat = 0.050 ± 0.002 s-1, Kcat/KM = 219 ± 3 M-1s-1)
